Restriction digests of the clone give the following sizes (kb): EcoRI--5.1 kb doublet; HindIII--10.25; PstI--10.25; EcoRI/BamHI--5.6, 4.1, 1.25; EcoRI/HindIII--5.1, 4.4, 0.9. The 0.9 kb EcoRI/HindIII fragment represents the HPRTP2 gene. The insert includes a 5.0 kb EcoRI fragment and a 0.9 kb EcoRI/HindIII fragment, with the latter subcloned from Hulambda5. The 3.8 kb constant band is from the X chromosome. Enzyme(s) not detecting polymorphism: TaqI, PstI, BstNI, XbaI, SacI, PvuII, HindIII, HincII, StuII, MboI, EcoRV. |
