The insert contains the following sites separated by (bp)(approx): SalI- 300- SacI- 200- BamHI- 800- BamHI- 375- SphI- 675- PstI- 450- HindIII. Restriction digests of the clone give the following sizes (kb): HindIII/SalI--2.75 doublet; PstI--3.4, 2.1; EcoRI--5.5; BamHI--4.4, 0.8, 0.4; XbaI--5.5. This insert is closely linked to the centromere of human chromosome 1. This probe detects a band of 7.4 kb in EcoRI-digested genomic DNA from human cell line HT1080 and in several human-hamster hybrids that contain an intact human chromosome 1. The insert was derived from a minichromosome which includes the centromere of human chromosome 1. |
