Animals were immunized with a synthetic peptide with the sequence YTLVREIRQYRLKKESKEEKTPGC corresponding to peptides 157-180 of the v-ras K oncogene. Spleen cells were fused with Sp2/0 myeloma cells. The antibody reacts with the synthetic v-ras K oncogene peptide. In ELISA testing, the antibody weakly detects the homologous region in the ras H sequence. In immunoblot testing a serum protein of approximately 45 kd is detected. The IgG1 and IgG2b reactivities of the antibody with the v-ras K peptide (157-180) was confirmed by ELISA. IgG1 was strongly positive while IgG2b was only slightly positive. |

Medium Renewal: Add fresh medium every 2 to 3 days (depending on cell density) Cultures can be maintained by the addition of fresh medium or replacement of medium. Alternatively, cultures can be established by centrifugation with subsequent resuspension at 2 X 10 exp5 viable cells/ml. Maintain cell density between 2 X 10 exp5 and 2 X 10 exp6 viable cells/ml. |
