| Disease |
malignant fibrous histiocytoma |
| Age |
adult |
| Applications |
The p53NiS1 cell line was derived in June 1997 by explant culture of a primary malignant fibrous histiocytoma.
The cell line demonstrates hypermethylation of the tumor suppressor p16, loss of heterozygosity of p53, and activation of the MAP kinase signaling pathway. |
| Derivation |
The p53NiS1 cell line was derived in June 1997 by explant culture of a primary malignant fibrous histiocytoma. The tumor developed three months after implantation of nickel sulfide (NiS) intramuscularly into the right rear leg of a p53 heterozygous mouse. |
| Oncogene |
p53 (deletion in exon 5); p16 -; p19ARF + |
| Genes Expressed |
p53 (deletion in exon 5); p16 -; p19ARF + |
| Tumorigenic |
No |
| Effects |
No, tumors did not develop within four months in nude mice (3/3) inoculated with 1 X 10 exp6 cells |

| Complete Growth Medium |
The base medium for this cell line is ATCC-formulated Dulbecco's Modified Eagle's Medium, Catalog No. 30-2002. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
|
| Subculturing |
Subcultivation Ratio: A subcultivation ratio of 1:4 to 1:5 is recommended Medium Renewal: Every 2 to 3 days Remove medium, and rinse with 0.25% trypsin, 0.03% EDTA solution. Remove the solution and add an additional 1 to 2 ml of trypsin-EDTA solution. Allow the flask to sit at room temperature (or at 37C) until the cells detach. Add fresh culture medium, aspirate and dispense into new culture flasks. |
