| Applications |
Ramos.2G6.4C10 cells provide a sensitive and specific assay system for IL-4 that is insensitive to the presence of other lymphokines and cytokines such as interleukins (IL-2, IL-2 and IL-6), colony stimulating factors (G-CSF, M-CSF and GM-CSF) and interferons alpha and gamma (IFN alpha, IFN gamma). Tumor necrosis factors alpha and beta (TNF alpha, TNF beta) cause only minor interference.
The assay is sensitive to IL-4 concentrations in the range 50 to 800 pg/mL. |
| Storage Conditions |
liquid nitrogen vapor phase |
| Derivation |
This line was derived from the Ramos (RA-1) cell line (see ATCC CRL-1596) by repetitive cloning with selection for low constitutive expression of CD23 and maximum stimulation of CD23 expression by IL-4. |
| Clinical Data |
3 years Caucasian male |
| Antigen Expression |
CD23 + |
| Receptor Expression |
interleukin 4 (IL-4); low affinity IgE (CD23) |

immunoglobulin (surface and secreted) |
| Comments |
Both the parental line (ATCC CRL-1596) and the derivative cell line Ramos.2G6.4C10 (ATCC CRL-1923) have about 1500 IL-4 binding sites per cell.

| Complete Growth Medium |
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, ATCC 30-2001. To make the complete growth medium, add the following components to the base medium: fetal bovine serum (ATCC 30-2020) to a final concentration of 10%.
|
| Subculturing |
Cultures can be maintained by addition or replacement of fresh medium. Establish new cultures at 5 x 10 5 viable cells/mL and maintain between 1 and 2 x 10 6 cells/mL.
Medium Renewal: Add fresh medium every 4 to 6 days (depending on cell density) |
