Restriction digests of the clone give the following sizes (kb): XhoI/PstI--1.8, 1.2, 1.1, 0.15; PstI--3.1, 1.28; XhoI--3.0, 1.35; BamHI--3.1, 0.65; HindIII--4.3. To obtain a full length (1.2 kb) fragment of the cDNA, cut with XhoI. To remove all vector sequences but 40 bp at the 3' end, cut with XhoI/PstI. The ApaI RFLP is distinct from the allelic isozymes because of different allele frequencies and because all 4 haplotypes of the 2 polymorphisms have been detected. |
