Insert contains the following sites separated by (bp) (approx): (SmaI)- 50- HinfI- 10- HinfI- 100- BglI- 80- SstI- 240- PstI- 50- SstI- 290- HinfI- 20- PstI- 15- (SmaI). Restriction digests of the clone give the following sizes (kb): PstI--3.2, 0.43; HindIII--3.7; EcoRI/HindIII--2.8, 0.84; EcoRI/BamHI--2.85, 0.82; EcoRI--3.7. Insert contains human beta-spectrin cDNA corresponding to an internal region of the protein from beta-9 through beta-12. A 284 bp SstI fragment and a 630 bp HinfI/BglI fragment were used to detect polymorphisms with PstI and PvuII, respectively. Detected a 7.8 kb message in RNA blots from two leukemia cell lines (K562 and HEL). Enzyme(s) not detecting polymorphism: several including BamHI and EcoRI. |
