Restriction digests of the clone give the following sizes (kb): BamHI/EcoRI--6.2, 3.4, 0.4; BamHI--6.4, 3.7; EcoRI--10.0; HindIII--10.0. The sequence is a gene for an extended-length structural mimic of the IIC10 peptide from the winter flounder (Pseudopleuronectes americanus) fused to the 8th codon of LacZ'. When the clone is digested with BamHI/EcoRI, the 0.3 kb band is the synthetic antifreeze peptide. For expression of the insert, the plasmid must be transferred to a host expressing T7 RNA polymerase such as JM109(DE3). |
