Restriction digests of the clone give the following sizes (kb): ClaI & EcoRI--3.4, 2.6, 1.8; HindIII--6, 1.8; XbaI--4.2, 3.6. AAV2 insert contains the following restriction sites (approximate kb from the 5' XbaI site): BamHI--0.85; BglI--0.35, 4.2; EcoRI--1.6, 1.8; HindIII--1.7; KpnI--1.7, 4.0; PstI--0.3, 1.75, 4.05; SacI--0.6; SalI--1.25; XhoI--2.05, 2.2. Helper virus for packaging helper-free recombinant AAV. Plasmid provides all trans functions (replication and capsid proteins) necessary for packaging AAV. Useful for creating recombinant AAV in conjunction with the cloning vector pAAV/neo (ATCC 68065). Constructed by replacing the inverted terminal repeats of the AAV2 genome with inverted repeats from adenovirus type 5. The poison sequence of pBR322 was also removed. The AAV portion of the insert can be excised by digestion with XbaI. |
