Restriction digests of the clone give the following sizes (kb): EcoRI--9.0, 4.6; BamHI--6.6, 2.2, 2.0, 1.8, 0.9, 0.62; HindIII--7.4, 5.3, 0.65, 0.48, 0.36; PstI--7.0, 3.65, 0.96, 0.87, 0.68, 0.37, 0.22; SalI--17.0. Codominant segregation of the BamHI RFLP demonstrated in 3 families. Detects genomic restriction fragments of the following sizes (kb): owl monkey HindIII--7.4, 4.6, 2.7; mouse HindIII--10.5, 4.0. Subcloned from lambda hINS-3. Contains 5' flanking sequences, exon 1 encoding part of the 5' untranslated region of IGF2, and part of the adjacent intron 1. Allele frequencies are different (A1 = 0.79, A2 = 0.21) in a Chinese population. The presence of the extra copy of pBR322 does not appear to generate any hybridization problems because the entire plasmid has been used as a probe. The construct appears to include 2 copies of the vector with the 8.6 kb insert from lambdahINS-3 because the intensity of the 4.4 kb band in the EcoRI digest is the same as the 8.8 kb band and there is an extra band in the SalI digest.
