Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.7, 0.5; EcoRI--3.2; HindIII--3.2. Distributed in aliquots of 200 ng dried. Constructed by inserting the SphI/HindIII fragment [intervening sequence (IVS) minus the first 44 bp] from pBGST7 (ATCC 40288) into pUC18, and adding a synthetic EcoRI/SphI fragment containing a T7 promoter and bp 22-44 of the IVS. After linearizing with ScaI, the T7 runoff transcript can be used to produce the ribozyme. |
