The insert contains the following sites (separated by approximate distances in kb): HindIII- 0.8- HindIII- 0.1- XhoI- 0.13- BamHI- 0.9- EcoRI- 1.6- HindIII- 1.3- EcoRI. Restriction digests of the clone give the following sizes (kb): SmaI/SalI--7.3, 6.3; SalI--13.7; EcoRV--8.5, 4.4, 0.87; XbaI--8.5, 5.2. A BglII (-820 nt from translation initiation site) to PstI (-72) fragment has been used to detect a mRNA of about 1.5 kb, presumed to encode mitochondrial fumarase. A 2.9 kb HindIII fragment detects 3 mRNAs, with the largest (about 1.9 kb) presumably a product of an unidentified open reading frame 3' to FUM1. The 1.35 kb mRNA is apparently the mRNA for cytoplasmic fumarase. |
