Restriction digests of the clone give the following sizes (kb): EcoRI--9.0, 5.8; BamHI--6.4, 5.6, 2.25, 0.7; PstI--14.8; BglI--9.4, 2.4, 1.1 (doublet), 0.8; HindIII--11.0, 2.0, 1.4, 0.4. In the strains that demonstrate amplification, variant bands are detected in Southern blots when genomic DNA is digested with MspI, HindIII, BglII, TaqI, BglI, BclI, MboI and EcoRI. This sequence exists in about 20 copies in strains A/J, AKR/J, BALB/cJ, CBA/J, C3H/HeJ, DBA/2J, SWR/J, and C57BL/6J (all on the X chromosome). In other strains, the number of copies is 80 - 100, with some of the additional sequences being on chromsome 11 (C57BL/10ScSn mdx, C57BL/10J), chromosome 17 (SJL/J, PO) or chromosome 13 (NZB/B1NJ). Hybridizes to the X chromosome of Mus caroli. Also detects multiple bands in the human genome. Isolated from a flow-sorted library enriched for the derivative X(7) chromosome produced by the Cattanach translocation T(X;7)1Ct. |
