The insert contains the following restriction sites (in approximate kb from the end of the insert closer to the 5' end of the gene): SstI--0.2, EcoRI--0.3, HindIII--1.0, 4.3; BamHI--1.05; PstI--1.2; BalI--1.4, 1.9, BglII--4.7, 5.3. Restriction digests of the clone give the following sizes (kb): BamHI--13.6; EcoRI--9.4, 2.3, 1.9; HindIII--5.4, 3.8, 2.1, 1.2, 1.1. Upstream flanking regions, starting from the HaeIII site at -322, contain the regulatory elements necessary for glucose-repressible transcription. HAP-regulated transcription requires sequences between a Sau3A site at -484 and the HaeIII (-322) site. Both a 675 bp BclI fragment from within the reading frame and a BamHI/PstI fragment from the upstream region detect 2 transcripts of 2.3 kb and 2.0 kb in approximately equal abundance. There are 2 start sites for transcription, -109 and -254. |
