The insert contains XbaI, PstI, and DdeI sites. The C to G substitution at nucleotide 374 changes the amino acid sequence from Cys125 to Ser125 and introduces the DdeI site. The amino terminal sequence is Met-Pro-Thr-Ser, with Met removed during processing. This lacks the native N-terminal Ala. The Escherichia coli trp promoter is contained in an EcoRI/HindIII fragment. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. |
