Growth Conditions
Temperature: 37°C
Recommendations for Infection: For best results, pre-treat cells with 25 µg/mL DEAE-Dextran (MP Biomedicals 02195133) for 30 minutes at 23°C. Prepare dilution of virus in minimum amount of volume (e.g. 1 mL per 25 cm2). Add virus dilution to host cells and rock or gently vortex every 15-20 minutes for the first 1-2 hours of incubation at 37°C. Re-suspend cells at an appropriate density. Incubate at 37°C in a humidified 5% CO2 atmosphere.
Incubation: 12-21 days at 37°C in a humidified 5% CO2 atmosphere, until CPE is progressed through 80% of the culture.
