Restriction digests of the clone give the following sizes (kb): PstI--4.5, 0.16 (doublet); EcoRI--4.8; HindIII--4.8; BamHI--4.8; AvaI--4.8; PvuI/NdeI--2.9, 1.8. The cryptdin mRNA (450 - 500 nt in length) is abundant in Paneth cells and can be detected in the testis and the brain. The gene maps to the proximal region of chromosome 8, within 1.28-21.27 cM of the Xmv-26 locus. Contains a open reading frame encoding a highly polar protein of 70 aa with all 6 cysteine residues in the COOH-terminal half. There are about 100 untranslated nucleotides 3', including poly(A). The insert lacks 115 bp of 5'-terminal sequence of the mRNA (which has a total size of 0.45 kb). This probe has been used to isolate several cryptdin-related cDNAs from an intestinal cDNA library. |
