The insert includes the following restriction sites (approximate kb from the 5' end): PvuII--0.18, 0.42; HindIII--0.64. Constructed from lambda 42-4 and lambda 16-4. The insert begins in the translated region and extends to the naturally occurring EcoRI site at nt 1070 in the 3' untranslated region. Contains the complete coding sequence for the mature protein, but lacks the 5' portion of the signal peptide coding sequence. Also contains part of the 3' untranslated region. A 578 bp AvaI/HindIII fragment encodes amino acids 8-157. |
