There are PstI sites at the 5' and 3' ends of the cDNA, and a PvuII site at position 1171 of the published sequence. Restriction digests of the clone give the following sizes (kb): HindIII/HincII--4.5; HindIII--4.5; PstI--2.8, 1.5 + smaller; EcoRI--4.5; HindIII/EcoRI--2.8, 1.9. Contains the complete cDNA. The insert contains pUC9 sequences at the 5' and 3' ends. The insert is oriented such that transcription from SP6 gives the sense strand and transcription from T7 gives the antisense strand. The HindIII site is at the 5' end and the HincII site at the 3' end. Enzyme(s) not detecting polymorphism: AvaII, BamHI, EcoRI, HindIII, MspI, PstI, PvuII, RsaI, TaqI. |
