Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--4.4; NotI--4.4; EcoRV--3.2, 1.2; PstI--4.4; EcoRV/NotI--2.6, 1.2, 0.6. Donor plasmid for gene disruption experiments in S. pombe. The AccIII ends can be partially filled in with dCTP using T4 DNA polymerase, and ligated to NotI ends partially filled in with dGTP. The vector was derived from pUC18 by replacing the HindIII/EcoRI MCS with an adapter providing a MCS with the following sites: AccIII EcoRV SpeI NotI AccIII. |
