Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.6, 1.9; EcoRI--4.6; HindIII--4.6; BamHI--4.6; HincII--uncut; SacI--4.6. This clone has demonstrated some instability. We recommend picking single colonies to identify the correct plasmid and amplifying with antibiotic selection. Codominant segregation was observed in 32 three generation families. RFLPS were observed after competitive hybridization with total human DNA. Insert is a 1.9 kb HaeIII fragment from cosmid cTBQ7 (isolated from somatic hybrid cell line 762-8A which included human chromosomes 10 and Y). |
