Restriction digests of the clone give the following sizes (kb): EcoRI--3.4, 2.4; EcoRI/BamHI--2.9, 2.3, 0.39; BamHI--5.7; PstI--5.7; PvuI--5.7. EcoRI detects a 2.6 kb band common to X and Y chromosomes. TaqI detects a 4.3 kb Y-specific band and X-specific bands of 3.2 and 1.5 kb. The HindIII/TaqI polymorphism has demonstrated co-dominant segregation in 8 families. The StuI polymorphism demonstrated Y-linked inheritance in 6 families. Wash in 2X SSC at 68C. |
