Restriction digests of the clone give the following sizes (kb): PstI--2.8, 0.34; BamHI--3.2; EcoRI--3.2. This clone is useful in hybridization tests for the detection of tobacco streak virus (necrotic shock) infection in Fragaria spp. Insert hybridizes with NS (necrotic shock) RNA 3 from strawberry and R RNA 3 from blackberry but not to R RNA 4 in northern blots of genomic RNA. Isolated from Fragaria vesca. cDNA was synthesized from virion genomic RNA, dC-tailed and cloned into the dG-tailed PstI site of pUC8. |
