Restriction digests of the clone give the following sizes (kb): NdeI--4.3; SphI--4.4; SacI/XhoI--2.9, 1.4. Insert contains the following restriction sites (approximate kb from the 5' end): BamHI--0.35; BglII--0.08; HindIII--0.07; NcoI--0.12, 0.24; NdeI--1.00; PvuI--0.10; SphI--1.20; XbaI--0.52. Encodes a truncated form of Vcam1 including Ig domains 1-3 and having a different carboxy terminus. The 5' end of the gene was generated by PCR and was ligated to the 3' portion of the clone N10 at the BamHI site. The resulting full length cDNA was confirmed by sequencing. |
