Restriction digests of the clone give the following sizes (kb): EcoRI--4.1, 2.9, 2.8; HindIII--7.4, 0.96, 0.8, 0.58; BamHI--3.8, 3.4, 2.0, 0.8; PstI--4.7, 3.0, 1.2, 0.8; AccI--4.4, 2.1, 1.18, 0.8 (doublet), 0.6; HincII--3.9, 1.7 (doublet), 1.3, 1.07; SalI--9.8. The insert contains the following restriction sites (in kb from the 5' end): EcoRI--2.7; HindIII--4.5, 5.25, 6.2. The complete coding sequence is included in the 5' EcoRI fragment of 2.7 kb, The 2.7 kb EcoRI fragment and the 1.8 kb EcoRI/HindIII fragment detect mRNA of 7.5 and 6.0 kb. The 3' 0.6 kb HindIII fragment detects only the 7.5 kb mRNA. The clone was derived from the 7.5 kb mRNA. which detects genomic EcoRI fragments of 2.1 kb (Zfa) and 1.9 kb (Zfx). The 1.8 kb EcoRI/HindIII fragment from the insert detects genomic EcoRI fragments of 8.2 kb (Zfa) and 5.3 kb (Zfx). |
