Restriction digests of the clone give the following sizes (kb): BamHI--7.0; HindIII--4.3, 2.7; XhoI--7.0. The 2.7 kb HindIII fragment contains the actin promoter and the partial actin genomic DNA. The 4.3 kb HindIII fragment contains the CAT gene, the SV40 splicing and polyadenylation sequences and the pUC18 vector. Contains the beta-actin promoter with its CAAT and TATA sequences, 1.5 kb of upstream flanking sequence, the transcription initiation signal, exon 1 (untranslated), intron 1, and a small segment of exon 2. SV40 promoter sequence deleted. |
