Restriction digests of the clone give the following sizes (kb): BamHI--6.0, 3.4, 2.8, 1.5, 0.6; EcoRI--7.6, 3.9, 2.5; BglII--4.4, 4.4, 3.5, 2.3. Contains the complete coding sequence (nt 100 to nt 8541), with the mature peptide encoded from nt 2389 to nt 8538. May be transfected into Chinese hamster ovary (CHO) cells in the ADA selective medium. The vector contains the SV40 ori, the adenovirus major late promoter and tripartite leader, the SV40 polyadenylation signal, the adenovirus VA I and VA II genes, murine adenosine deaminase (ADA) cDNA sequence, a DHFR cDNA sequence and pBR322 sequences. |
