Restriction digests of the clone give the following sizes (kb): AvaI--4.6, 2.8; SalI--7.4; PstI--5.4, 1.49; HindIII--5.4, 1.95; EcoRI--5.4, 1.73. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture. The combined heterozygosity of the polymorphisms detected by p13-1-82 (ATCC 59004, 59005) and p13-2-21 (ATCC 59014, 59015) is 72%. Seven of 24 possible haplotypes occur with frequencies of 3-38%. The MspI (A alleles) and the TaqI (C alleles) polymorphisms are at the same locus (alpha locus). This polymorphism is also detected by p13-1-25 (ATCC 59870). p13-1-82 (ATCC 59004, 59005), p13-2-31 (ATCC 59014, 59015), and p13-1-25 (ATCC 59870, 59871) were subcloned from cosmid 1-13 (C1-13). |
