Restriction digests of the clone give the following sizes (kb): BamHI--4.6, 2.7; BstEII--7.4; SstI--7.0, 0.4; NotI--7.4; EcoRI--5.0, 2.8. Digestion with EcoRI produces fragments of 5.0 and 2.4 kb. Bacteria may grow slowly due to the high plasmid copy number. Encodes a chimera in which aa 1-48 of processed human factor VII (Gla domain), with mutations A1Y, K32E, D33N, A34T, K38T, L39E replacing the corresponding region of factor IX. |
