Restriction digests of the clone give the following sizes (kb): NdeI/SalI--2.3, 0.7, 0.3; HindIII/PstI--2.0, 0.8, 0.45; PvuI/SacI--2.3, 0.9. The vector uses a polycistronic expression system designed to reduce formation of secondary structure of the mRNA. The promoter is followed by a ribosome binding site (RBS), the N-terminal salmon growth hormone coding sequence, a second RBS, in frame stop codon and bovine growth hormone coding sequence. |
