Restriction digests of the clone give the following sizes (kb): BamHI/XhoI--3.0, 1.7; EcoRI--4.4, 0.3; PstI--3.3, 0.7, 0.5, 0.3; PvuII--2.5, 1.6, 0.65. The insert contains the following restriction sites (approximate kb from the 5' end): EcoRI--0.28, 0.40; PstI--0.43, 1.10, 1.36; PvuII--1.26. Excising the insert with BamHI/XhoI will add approximately 20 bp from the MCS of the vector sequence to the 5' end of the insert. |
