Restriction digests of the clone give the following sizes (kb): PstI--6.6, 4.6, 3.6, 1.5, + smaller; HindIII--9.4, 6.6, 1.7; EcoRI--6.0, 4.0, 2.7, 2.4, 1.5, + smaller; BamHI--16.5, 1.1; SalI--17.0. Codominant segregation demonstrated in 49 3-generation families. Detection of the MspI polymorphism (alleles 1.6 and 0.6 kb) was verified by CEPH. Prehybridization and hybridization should be carried out in the presence of 250 ug/ml total human DNA. RFLPs are observed under otherwise normal conditions of hybridization and washing. Enzyme(s) not detecting polymorphism: BamHI, BglII, EcoRI, HindIII, PstI, PvuII, RsaI, TaqI. |
