Restriction digests of the clone give the following sizes (kb): HindIII--6.8, 2.7; SstI--2.7, 2.2, 1.9, 1.25, 1.1, 0.2; BglII--9.4; PvuII--2.3, 2.1, 1.65, 1.5, 0.78 (doublet); EcoRI--9.4. The insert contains the following restriction fragments (approximate kb from the 5' end): SstI--0.1, 1.14, 2.3, 4.4; BglII--2.8; ApaI--2.0, 2.5, 3.14, 5.3. Contains the complete gene (2 exons and 1 intron) and 5' and 3' flanking sequences. The 500 bp ApaI fragment from the promoter region is sufficient to confer responsiveness to tetradecanoyl phorbol acetate in transient transfection assays. |
