Restriction digests of the clone give the following sizes (kb): HindIII--20.0, 15, 3.7, 3.6, 0.5; KpnI--19.0, 17.0, 3.9, 2.9, 1.9, 1.6; PvuI--23.0, 12.5; SacI--20.0, 9.4, 8.0, 4.4, 2.9, 1.5, 0.8; SacI/EcoRI--20.0, 9.4, 6.6, 4.0, 1.3, 0.9, 0.6; XhoI--20.0, 18.0, 9.0. The insert contains the following restriction sites (approximate kb from the 5' end): SacI--4.6, 12.0, 14.8, 16 (polylinker site); HindIII--3.0, 3.5, 7.2, 10.8. A 1.2 kb SacI/EcoRI fragment containing the Vgamma3-Jgamma2 rearranged gene (S12SR, ATCC 59590, 59591) has been used to detect rearrangements. Contains a productive rearrangement of Vgamma3 and Jgamma2. The insert begins about 0.5 kb upstream of Vgamma2 and extends 2.5 kb beyond exon 2R of TCRGC2.
