Restriction digests of the clone give the following sizes (kb): BamHI--3.0; BglI--3.0; EcoRI--2.7, 0.25; DraI--2.20, 0.70; HindIII--3.0. A PCR product corresponding to the unique sequence beginning at nt 901 of GenBank M22958. The 3' BamHI site in the polylinker was removed after cloning. Transcription from the SP6 promoter gives the sense strand (linearize with HindIII). Transcription from the T7 promoter gives the antisense strand (linearize with SmaI). |
