Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.4, 0.23; SmaI--uncut; EcoRI--2.7; BamHI--2.7; HindIII--2.7; EcoRI/BamHI--2.5, 0.2. The antibodies encoded by myeloma MOPC-167 are specific to phosphorylcholine. The insert was subcloned from ChM167alpha10.1. A 210 bp fragment can be excised by an EcoRI/BamHI double digest, which cuts in the polylinker. The vector pBSc was constructed by inserting the 382 bp PvuII fragment from pBluescribe (Stratagene) into pACYC184 (pUC8CmR) which had been partially digested with PvuII. |
