Restriction digests of the clone give the following sizes (kb): EcoRI--5.0, 2.6; BamHI--7.6; HindIII--7.6; PstI--4.0, 1.4, 1.1, 1.0; EcoRI/HindIII--5.0, 2.6. A high stringency wash (0.1X SSC, 0.1% SDS, 65C) is required because of significant lane background. Pre-hybridization of probe with sheared total human DNA is useful. This EcoRI fragment was isolated from phage LAMP92. Enzyme(s) not detecting polymorphism: EcoRI, HindIII, MspI, BamHI, PstI, StuI, RsaI, XbaI. |
