Restriction digests analyzed on agarose gels give the following sizes (kb): EcoRI--4.7, 3.2; BamHI--7.8. A general purpose integrating Saccharomyces cerevisiae/Escherichia coli shuttle vector. A 1.1 kb HindIII fragment containing URA3 and a 2.2 kb SalI/XhoI fragment containing LEU2 were cloned into the HindIII and SalI sites, respectively, of pBR322. Unique restriction sites are in the 3' untranslated regions of URA3(SmaI) and LEU2(SalI). Orientations are such that transcription from both URA3 and LEU2 is clockwise. |
