Restriction digests of the clone give the following sizes (kb): BamHI--25.0, 12.5, 2.0; SalI--19.5, 14.5, 10.0. phi6-3 contains repeated sequences and must be prehybridized with total human DNA before it will detect single copy bands. Standard hybridization and washing conditions can be used, with a final wash in 0.1X SSC/0.1% SDS at 68C. MspI detects 2 diallelic RFLPs. The constant band at 2.5 obscures the B2 allele when the entire phage is used as probe. Because the 2 loci are in total disequilibrium, use of locus A alone provides all available information. |
