Restriction digests of the clone give the following sizes (kb): BglII--uncut; ClaI/BamHI--3.4, 2.8, 0.5; EcoRI--6.6; BamHI/PvuII--6.0, 0.5. URA3d is a modified URA3 gene which is not efficiently expressed and requires a large number of plasmids to complement a ura3 mutation. Shuttle expression vector. Permits expression in both S. cerevisiae and mouse cells. The EcoRI, SmaI, and BamHI sites are 3' to the unique available in-phase initiation codon under control of the SV40 early promoter. Constructed by inserting a 1.207 kb ClaI/BamHI fragment containing URA3d into pCYM2 cut with BglII and ClaI (nt 3469 and 3589 according to the pCYM0 numbering scheme). |
