Restriction digests of the clone give the following sizes (kb): XbaI/BamHI--2.5, 0.75, 0.5; HincII--2.8, 0.85; BglII--3.7, HindIII--3.7, BglII/BamHI--2.2, 1.5. The XbaI site is upstream of the cDNA insert so excision with this enzyme will include some vector sequence. A new deposit was made for this clone. It is recommended that ATCC 68636 be used instead of ATCC 68507. Contains 1000 bp of the coding region of mature hIL-6 plus 445 bp of the 3' untranslated sequence. Because the host expresses T7 RNA polymerase, and the cDNA is inserted downstream of the phi10 T7 promoter, this strain shows high levels of expression. The product is the mature protein and does not require in vitro cleavage of a fusion protein. |
