Restriction digests of the clone give the following sizes (kb): PstI--4.3, 0.8, 0.3; EcoRI--uncut; PvuII--5.6; BamHI--5.6; EcoRI/PvuII--5.6. The insert contains an internal PstI site (two fragments of 787 and 214 bp). The insert encodes TF from amino acid 403 to COOH terminus with 166 nucleotides of 3' untranslated region. Incomplete dominance for the PstI RFLP has been demonstrated in one informative family. HindIII variant shows amplification of TF sequences. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. Enzyme(s) not detecting polymorphism: EcoRI, PvuII, BamHI, TaqI, MspI. |
