Restriction digests of the clone give the following sizes (kb): SstI--5.2, 3.0; EcorI--4.4, 3.8; HindIII--6.6, 1.6; BamHI--8.2; XbaI--6.0, 2.2. The insert includes the following restriction sites (in approximate kb from the 5' end): EcoRI--0.2; HincII--2.3; XbaI--3.0; BglII--3.3. The insert includes exons 1, 2, 3 and upstream and downstream sequences. Exons 1 and 2 can be excised by a HincII/XbaI double digest yielding a 750 bp fragment. The genomic copy number for the entire insert is not unique. There is a repetitive element located within the third intron. The HincII/XbaI fragment does detect a unique DNA fragment. Transcription from the T3 promoter gives the sense strand. |
