Restriction digests of the clone give the following sizes (kb): BamHI--3.6, 1.9; HindIII--3.7, 1.8; BglII/HindIII--3.7, 1.1, 0.7; PvuII--4.4, 1.1. The insert contains the following restriction sites (approximate kb from the 5' end): PvuI--0.15, 0.21; HindIII--0.92; EcoRV--1.41. Contains the complete coding sequence (nt 31-567). The mobility of the protein produced by COS-7 transfectants is not affected by tunicamycin. The 3' untranslated region contains an Alu repeat. A 0.7 kb BglII/HindIII fragment from the insert is repeat-free. The vector is modified from pcD, with a promoter constructed from the SV40 early promoter and the R segment and U5 sequence of the long terminal repeat of human T-cell leukemia virus type 1 (HTLV-1 LTR). |
