Restriction digests of the clone give the following sizes (kb): PstI--2.6, 2.4 (partial); HindIII--3.4, 1.5; BamHI--3.2, 1.35, 0.35. The insert contains the following restriction sites (approximate kb from the 5' end): BamHI--1.2, 1.55; BglII--1.0; ClaI--1.7; EcoRI--0.1; EcoRV--1.95; HindIII--0.75; KpnI--1.8; NcoI--0.3, 1.45; SalI--1.4; SmaI--0.6, 1.6. Virus was isolated from cucumber, Cucumis sativus. The insert hybridizes to the genomic length RNA (4.26 kb) and two shorter RNAs of about 1.9 kb and 1.6 kb. |
