Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--2.8, 2.0; EcoRI--5.5; HindIII--3.05, 1.9. PstI, MspI, TaqI, and PvuII identify the same polymorphism. Codominant segregation demonstrated in 55 3-generation families. The probe was pre-associated with excess human DNA before hybridization. Otherwise, RFLPs were observed under normal hybridization and washing conditions. Enzyme(s) not detecting polymorphism: BglII, RsaI. |
