Raccoon poxvirus CDC # V71-I-85A was plaque purified three times then plasmid pKB3-JE-13 (ATCC 40280) was transfected to cause insertion of CVS rabies glycoprotein cDNA in the thymidine kinase gene, inactivating it. Plaque titration is on CV-1, BSC40 or Rat2 cells. Adsorb virus for 2 hours at 36C, overlay with DMEM + 2% FBS (liquid overlay is OK) incubate for 72 hours, stain monolayer with 0.01% crystal violet in 10% formalin, rinse with PBS. |
