Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--6.6, 4.4, 4.1, 2.5, 2.3 (doublet), 2.1, 1.9 (doublet), 1.7, 1.6, 1.3 (doublet), 0.9 (doublet), 0.8, 0.6 (triplet), 0.5 (doublet), 0.35. Reported to contain EcoRI/HindIII fragments of the following sizes (kb), ordered as in the genome: 1.267, 0.937, 0.790, 0.485, 2.085, 1.906, 1.533, 0.429, 1.983, 1.295, 4.392, 4.113, 0.891, 0.609, 6.113, 2.390, 0.486, 2.593, 2.368, 1.630, 0.606, 0.571. IMPORTANT: To prevent amplification of a rearranged and/or deleted cosmid, we recommend streaking on LB + amp plates at 30C and picking small colonies for liquid culture.
